Where was the first Spanish colony founded?

If we mean the colonial system of Spain in America, then the first colony is considered to be Santa Maria la Antigua del Darien. It was founded on the Isthmus of Panama in 1510. The colony was primarily used as a base for further advancement on the continent. In particular, the expedition of Vasco Nunez de Balboa came from here (he founded a permanent settlement).
