Why did Russian-Turkish relations become the main focus of foreign policy?

Several factors:
The Crimean Khanate, a vassal of Turkey (Ottoman Empire), the raids of the Crimean khans demanded that the Moscow kingdom gradually move south in the 17th century, a clash with Turkey became inevitable (the war of 1676-1681 and the Azov campaigns in 1695-96 and the war of 1735-1739)

18th century – access to the Black Sea, the construction of a fleet in the south of the country (started by Peter I, stopped in 1714, again under Catherine II)

XIX century – the desire of the Russian Empire to patronize the Orthodox population of the Balkan Peninsula (Serbs, Bulgarians), the desire to establish control over the Bosphorus and the Dardanelles and take Istanbul, which is the former Christian Constantinople.

Thus, a combination of several factors: religious-political, military (protection of the southern borders), economic (trade, development of fertile lands)
